Image analysis techniques such as chlorophyll fluorescence, UV-MCF multispectral fluorescence, thermography, and reflectance spectra represented by the NDVI normalized vegetation index are the most advanced and important non-destructive plant phenotypic detection and analysis techniques, especially for Studies on the detection, prediction and response mechanisms of various biotic and abiotic stresses in plants.

Jiangsu Academy of Agricultural Sciences used this modular plant phenotypic analysis technique to rapidly detect the mung bean insect pest and mung bean leaf spot disease, and quantitatively identified the pest resistance of different strains of mung bean.
Through the image threshold segmentation function of chlorophyll fluorescence imaging, the mung bean leaves infected with leaf spot disease were divided into non-infected parts, mild infected parts, moderately infected parts and severe infected parts according to the preset criteria. The evaluation and identification of leaf resistance is achieved by the difference in leaf distribution and area.
After being infected by bean image, due to the difference between the temperature of the insect body and the temperature of the uninfected mung bean, it is possible to quickly identify the infection of the bean image from the thermographic image.
Because the bean image infestation activates the defense mechanism of mung bean itself, the normal biochemical process is disrupted, and a large number of secondary metabolites are synthesized. UV-MCF multispectral fluorescence imaging is the best tool for non-destructive quantitative detection of secondary metabolic levels. In the figure below, there are different strains of mung bean control group and bean-infected treatment group. The leftmost column is a mung bean mutant, and the white spot on the surface of mung bean is bean beetle egg. Through the multi-spectral fluorescence imaging on the right side, it can be clearly seen that the bean-like infection significantly increased the secondary metabolism level of mung bean, and the background secondary metabolism level and resistance between different strains of mung bean were significantly different.
Jiangsu Academy of Agricultural Sciences hopes to use this research to further establish a rapid phenotypic identification system for mung beans and guide the breeding and production practices of fine varieties.
